I've taken glucosamine for about six months now. I took it when I started really getting into running because I was having some knee pain. I haven't had the knee pain since then, but I also started some different stretches, had my stride analyzed, bought some better shoes and worked on my form a little bit, so I can't really say that it's the glucosamine doing it. I'm afraid to quit taking it though, in case it is the reason I've been pain free. I figure it can't hurt to take it.
